Biological Function of Z-DNA I would try to have the citations linked through a subscript in the citation maker to quickly access the sources used. The use of a specific gene pathway was extremely helpful in understanding the biological function of Z-DNA. This portion of the writing was extremely well-written and the grammar seemed to be correct. In addition, the paragraph flowed really well in explaining the different biological functions of Z-DNA. I would perhaps also make some general statements about Z-DNA function for the casual reader who comes across this page, but this page seems to be understandable from a scientific standpoint. Biological Significance The ideas and facts presented in these articles and written portion seem directly relevant to the biological significance of Z-DNA. The use of these two specific diseases helps the reader to understand the importance or link of Z-DNA to Alzheimer's disease and Systemic Lupus Erythematosus. I would include hyperlinks to the Wikipedia pages of these two diseases so the casual reader has ease of access to learn about the diseases in general, especially Systemic Lupus Erythematosus. Also for the sources, I would add them through the Wikipedia citation editor so the sources will also be linked and can be easily accessed by clicking on this superscript number after the citation. A possible figure would be helpful for showing the link/mutations or pathway to this specific disease, but you made a great job of stating that it is unknown about a direct effect on the two biological conditions mentioned. I found another source which dealt with the regulation of Z-DNA structure of a specific gene, ADAM-12, which displayed some relevance in the reduction of breast cancer in vivo and in vitro.Cite error: There are <ref> tags on this page without content in them (see the help page). http://cancerres.aacrjournals.org/content/73/2/736.full-text.pdf

This would add one more possible biological significance for Z-DNA and hits on cancer, which was not mentioned often in the original Z-DNA Wikipedia article. Overall, I found this section to be extremely well-written and thoroughly detailed. You did an awesome job of explaining things in a clear and concise manner that is understandable to both the scientific community and the general public. The two biologically significant diseases were very notable, as it continues to be difficult to find more connections with Z-DNA to disease, due to the lack of clear linkage in research. Finding sources on this biological significance seems to be extremely difficult, but the sources are from reputable journals. Pathway Formation of Z-DNA from B-DNA

This portion of the research regarding Z-DNA seems to be extremely thorough and understandable to the reader. I would include images from pymol or possible images from online articles to enhance this topic of high-affinity binding proteins. I would make the same adjustments regarding citations as the previous paragraphs. I would try to find an image of this B* transition state or the binding of hZaADAR1, as this seems extremely important to the pathway of formation for Z-DNA. However, I found this section to be well-written, grammatically correct and learned a great deal about this pathway and its importance.

Overall

The group did an awesome job on writing and researching for this article. I hope some of these points can help, but the main ideas and important points all seem to be there. The use of images either from pymol or online sources might really enhance some portions of the writing, if possible. The sources used were great and seemed to be highly notable and significant. I would make those adjustments with citations to follow the typical Wikipedia style. I came away from reading this sandbox having learned a great deal about Z-DNA and its importance through both a chemical and biological standpoint. A lead section that is easy to understand- Might consider adding some more general information about z-DNA before diving into the specifics. The lead section of the current article also lacks a simple introduction to z-DNA so it would be valuable to add that. A clear structure- The headings for both sections are very similar. Its not immediately clear what the difference between "biological function" and "biological significance" is. I would either change the subheadings or merge the two under one subheading. Balanced coverage- Good Neutral content- Good Reliable sources-Good

A little bit of background on the EL3 gene (what it can cause) would help me better understand the significance of it.

“A study conducted on normal, moderately affected with Alzheimer’s disease, and severely affected with Alzheimer's Disease human brains showed the presence of Z-DNA in the DNA of those severely affected [1].” This sentence is a little confusing try rewording it.

Not sure why there are separate citation sections for each subheading. Maybe this will change automatically once its added to the real article.

There are a few grammatical errors and places that could be more concise.

I loved the pathway section! Lots of detail and fairly easy to understand.

Overall a lot of good contributions!

Biological Function The first sentence: "Z DNA binding" by? What is binding? You should provide an introductory statement.

Two critical components to the EL3 gene are the N terminus and the C-terminus. Is it critical component of the gene or the protein?

Should mention the name of the researcher at MIT-- was that also Rich?

Although you cite results, the story does not come across. You need to explain some of the results and its importance to Z-DNA.

Biological Significance "its presence"-- be specific. Presence where? in a gene? promoter? Is it the presence of Z-DNA or a sequence that forms Z-DNA? How did they show the "presence" of Z-DNA? What DNA was moderately affected? The entire genome? How was it shown? Why was it concluded that the transition is important. Provide more detail.
