Hi, I don't have a preference but we can keep the discussion and other assignments here on my sandbox if that's ok with you? I'll start making bullet points for the references I found and move these into my sandbox page (which will now be our groups sandbox page). Preliminary Outline

Hi, I added a section for the preliminary outline in our sandbox page. I started adding headings and information under the headings that we might need to write about in our contribution to the article. If you see anything that needs to be edited please do so, we can work on this together like we did for the references. If you want to move some headings around so that the article will flow better, please do. What I wrote is just to see the information we have on the computer so later we can move it around so that the flow of the article makes sense. I hope that made sense in itself :) Talk to you soon! Hey, I think this is coming along nicely. I changed and added a bit under the Mechanism section which I hope makes sense and flows well from initiation to termination. I felt like each should be its own subsection but if you don't like this we can discuss other ideas. I intended on looking for a few more refernces but with what you added today I think we have a great start with plenty of information. I also really like that pic of the replication fork. I could not find a good one as the one in WikiCommons had some disputes with it. I like how you added the subsections to the mechanism. It would make sense to make subsections instead of listing everything applying to the mechanism all in one paragraph. How do you think we should post this on the article talk page for other wiki editors to see? Do you think we should write out a paragraph explaining what we would like to contribute and then give our outline or just an explanation paragraph? I do agree that this is becoming a fun assignment! I can write a summary paragraph of what we have included so far. I think maybe the outline would be too much. Article Talk Page Summary

In looking for ways to improve this article it appears that there needs to be some serious restructuring. We propose to begin with a lead section to include an overview on location of replication in the cell and in the cell cycle and the importance of complete and precise replication of all DNA only once as errors can lead to incorrect links between daughter chromosomes, chromosome breaks or loss, or DNA lesions. This should then lead in to the need for and challenges faced by multiple origins of replication. At this point the article can then branch into a more detailed discussion regarding the mechanisms of DNA replication. Here we intend to begin a new section entitled Mechanisms of Replication with subsections labeled: Origins and Initiation, Replication Fork, Regulation and Termination. Topics to be covered in the origins section would include pre-RC assembly to entail ORC, protein requirements (Cdc6 and Cdt1), DNA helicase and Mcm2-7 complex. This section will also discuss the need for kinases and polymerases α, δ, and ε for the formation of the replication fork. The replication fork section will encompass leading and lagging strand inforamtion and other important components such as DNA gyrase, SSBs, and sliding clamp/loaders. The regulation section will go into a bit more detail regarding the location in the cell cycle and the various checkpoints and progression through these checkpoints. And finally termination will detail telomere regions and telomerase. This would provide better development of this topic and would flow better. Initial Contributions

Hello Autumn, I hope you had a great break. I wanted to touch base to see exactly how we would want to go about this Week 8/9 assignment. I was thinking we could break the work up a bit into sections and each intially tackle a section. We could then modify as needed. For instance since you had a good bit of information on the opening section maybe you could work on that section. I could then work on the Origins and Initiation, Replication Fork, and Termination subsections of the Mechanisms section while you could maybe take the Regulation section. In general I was thinking that I could have contribtutions done by the beginning of next week so that we could modify as needed before Thursday. I think that's a great idea to break up the work load, eukaryotic DNA replication is such a large topic! So I'll take on the lead section and regulation, but if you need help in the other areas let me know. Do you think we should leave the section about where in the cell this takes place and how important replication of the entire cell genome is to the cell and then lead off with the origins of replication and so forth, or do you think the article would benefit if we begin discussing the different mechanisms and then discuss where they are located towards the end? And I agree with by having our contributions done by the beginning of next week so that we can modify. We also have to write a summary of what we did by the next due date. Talk to you soon! I was just thinking that regulation of eukaryotic DNA replication goes along with every step of the whole replication process, so maybe I should tackle the Origin of replication and the pre-initiation complex and flow into the replication fork before replication begins and then you can take the process of replication and the termination or visa versa? This would work just as well. Do you think maybe I could include the location in the cell cycle in the Regulation section? I will say that I really like the way the DNA Replication article flows however I think there is some room for improvement in this section such as discussing how the previous Origins and Initiaton fit into the G1 phase and then how activation of the replication fork and replication process would begin with entrance into S-phase. I think the Regulation section could end up being a very good overview and wrap up of the process as a whole and could tie the concepts together really well although I dont't want to reiterate the information too much. Now I see where you are going with the flow of the article when comparing to the DNA replication article, this would be a good guideline as to how to incorporate all this information. I do think that a regulation section would be nice to have and we could probably incorporate what the previous editor added in regards to the cell cycle and the regulations there. I'm going to start adding information and when we both are done contributing for this unit section then maybe we can discuss the order of things if its not what you would want. Makes great sense as I think once we get the article laid out a bit better we may realize that it could stand some adjusting. Hello! I think we got a lot done with our initial contributions to the article. I read through what you added and it's written really well and flows nicely. Good work!! Can you take a look at what I wrote. I was wondering if the different paragraphs for each protein complex was too much? Also, we need to begin writing a Unit 9 Progress Report of what we contributed to the article on our sandbox page I believe, so I'm going to begin that now and you can add to it whenever you have time, and please revise anything that looks wrong to you for our assessment and article contributions. I think what you have looks great! Well written and very detailed!! I am trying to find some more scientific articles to be able to use for the Termination and Regulation section. Most of what I have came out of two textbooks so I don't have any references addded in yet. Let me know if you think this is a problem. Also do you want me to go ahead and throw in some replication fork information? I am not really sure if you had wanted to do this or if you would like me to take care of it. Let me know and I am happy to add in a little info for now. Also do we want to remove the previous information from location in the cell cycle down? I think this is alittle confusing but didn't want to remove it prematurely. I don't think using textbooks is a problem as long as you find some references it should be ok. If you would like to start on the replication fork that would be awesome! As for the information from location in the cell cycle I do think its confusing and a lot of the information there coincides with what you have written, so if you think its repeating information you have I would just take it down, or maybe take parts of it down? Take a look at the rearrangements that I did on the article. I think it flows better now. I changed the title of mechanisms to initiation and I think that with the rest of the article laid out the way it is this makes more sense. Hopefully you like it but don't hesitate to say otherwise...I will not take offense! Also I intend to add more to the replication section but it may not be before the deadline for the initial contributions. I think we have already tremendously improved this article but if you feel the replication fork is needed prior to tomorrow let me know and I can try to find some time to squeeze this in. I think changing the title of mechanisms to initiation works much better and flows nicely and is easy to understand where the article is heading. I think we have more than enough for our first contributions, but if you have the time to add more, the better. But I really do think we are in good shape. Can you take a look at our Unit 9 Progress Report on our sandbox group page? I'm kind of at a loss as to what to add and what our professor might want us to add. Thanks!!! I am glad we are pretty in sync with this project. I am really enjoying it so far! I am going to add a bit more to the progress report today. You can look it over this afternoon and add what you think may need to be included. I think so far the article lay out and your sections are really complete. Unit 9 progress report

Hi! I saw that you added our progress report to our groups page for the article, but do you think its supposed to go on our sandbox page that we chose to add all our information on? I could be wrong and I'm a little confused. Also, here is what the directions are for the report:

"Write a very brief progress report on your group page, in a new section titled "Unit 9 progress report". As a guideline, this should be a bulleted list of up to five items, with one or two lines per item, highlighting specific contributions that we otherwise might miss. For example, don't include "Wrote the introduction section", because that kind of contribution will be obvious. Do include things like "Used Inkscape to make the figure 'small nucleic acid lipid particle', and uploaded it to Wikimedia Commons", or "Updated the infobox-disease with several external links." If all of your contributions were prose, then your progress report might consist of only one line, "All of our contributions were prose.""

I am so sorry I missed that entire section of instructions for the summary. However I do believe we should put it on our group page. What I will do is delete what I have entered on our group page and redo this but keep the bulleted points on the sandbox page that we are working in. How does that sound? I think what you have written as a progress report looks great and after looking at what Dr. Ogg is expecting this is right on target. Eukaryotic DNA Replication Proteins List

I wanted to let you know that I added a list to the bottom of our eukaryotic DNA replication article that lists all proteins that we have mentioned or may mention in the bulk of our contributions. I thought this could be helpful for users who want to have a quick reference for the proteins involved in eukaryotic DNA replication and their functions. Hello- I have added a bit to the elongation and replication fork sections. Please feel free to check it out and let me know what you think so far. Also I was reading back through the suggestions that were posted previously on the articles talk page and I am not so sure if I agree with the logic of adding comparisons to prokaryotic DNA replication. My reasoning is that there is the main DNA replication article that gives a good overview of this already and I feel that our contributions should just focus more on eukaryotes. Hello-I saw this today. It's coming along nicely. I moved the replication checkpoint proteins under the regulation section as I think it may fit well there. Let me know what you think. I will also add geminin to the proteins list as well as a section briefly comparing prokaryotic and eukaryotic DNA replication. I like where you moved "replication checkpoint proteins", it makes more sense there. Geminin is already on the list, its towards the top, but look to see if the description makes sense for the protein. I'll start reading the article for grammatical errors and things like that. Hi! Everything looks so good, I think the section you added compares the basic differences between the two, which is good because I think if we went into any further detail then we would writing a whole new article on prokaryotic DNA replication. We could add a small chart that lists the proteins used in prokaryotic DNA replication to compare to eukaryotic. I can do this if I have some free time here at work. Just a thought.
