The flow cytometric approach to differentially stain RNA versus DNA with acridine orange.[1]. This method was the first to identify non-cycling (quiescent; G0 cells) and recognize other compartments of the cell cycle, is still being used in cell biology.[2]
Another widely used [3] methodology is (the TUNEL assay)[4][5]
The TUNEL assay made it possible to detect DNA fragmentation in nuclei of sperm cells of men with decreased fertility[6] This methodology, recognized by WHO, serves as the means of identification of sperm cells abnormality and detect infertility.[7]
The Fluorochrome Labeled Inhibitors of Caspases (FLICA assay) which detects caspase activation during apoptosis in relation to the cell cycle phase,[8] has found wide application in biology and medicine[9]
Still another methodology that found numerous applications in cell biology and medicine[10]to detect DNA denaturation made use of the metachromatic properties of acridine orangeto detect denaturation of DNA in individual cells.[11].
The above methodology when applied to sperm cells detected abnormality of chromatin structure of these cells associated with infertility[12]This assay, in conjunction with the TUNEL assay is used as the key method to assess male fertility in husbandry and human clinic[13]
The author pioneered in developing flow cytometric approaches with phosphospecific antibodies to detect phosphorylation of nuclear proteins such as pRB, histone H3 (serine10)and histone H2AX (γH2AX) in relation to the cell cycle phase[14][15]"
